Seeking to set the “Reply-to email address” above via the api. Appears it would be set via the endpoint:
https://a.klaviyo.com/api/campaign-messages/{id}/
Per below documentation (snippet of relevant object included), there is no key/value available to do so:
https://developers.klaviyo.com/en/reference/update_campaign_message

At this time, it’s not possible to update this field via API. However, I will forward this to our Product Team and update the thread if there are any changes in the future.
